Not at all. All the midpoint formula for x does is to take the average of two end point x values. It matters which is considered the leftmost and which is the rightmost.
Similarly, order is irrelevant in the midpoint formula for y.
Only when two points are being used to compute slope does 'consistency matter; if the order of computing the difference in x values is not the same as the order in computing the difference of y values, one will miscompute the slope.
